- RNA binding protein Fused in sarcoma (FUS) undergoes liquid-liquid phase separation (LLPS), resembling oil in vinegar. Evidence suggested that phase separation of FUS is crucially involved in amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD) pathogenesis and RNA regulates its droplet formation, suggesting that antisense oligos (ASOs) modulating FUS LLPS could be a promising drug candidate against ALS/FTLD. My efforts are designed to develop oligonucleotide drugs targeting phase separation for ALS/FTLD using a combination of biochemical and cellular technologies including iPSC-derived motor neurons and animal models.
